Job Description

Are you an ambitious researcher looking for your next challenge? Do you have an established background in structural mass spectrometry? Do you want to further your career in one of the UK’s leading research intensive Universities, in the world renowned Astbury Centre?

We are seeking a highly motivated and talented individual interested in undertaking an exciting 2-year postdoctoral project (with potential for an extension) in the laboratory of Professor Frank Sobott (Biomolecular Mass Spectrometry). You will make leading contributions to two funded projects, as well as having the opportunity to develop an own research profile by pioneering structural mass spectrometry approaches for nucleic acids.

Project 1 is BBSRC-funded in collaboration with the University of Bristol, with the overarching goal to study structure-function relationships of protein systems which repair DNA (“machines on genes”). This project specifically focuses on the human DNA repair factor CtIP which is an important hub protein within the human resectosome. Perturbations to this molecular assembly impact directly on DNA break resection, and these changes (and therefore DNA repair pathways) can be regulated by post-translational-modifications or disrupted in disease states.

You will use biochemical and biophysical methods to analyse the architecture and interactions of CtIP within the resectosome, including how these are affected by PTMs and disease-states. You will also, in collaboration with Bristol, assess the impact of these interactions on DNA break resection and its regulation. You will investigate these complexes – both protein and nucleic acid components – using state-of-the-art structural mass spectrometry (e.g. HDX-MS, hydroxyl footprinting, crosslinking and native and ion mobility), biophysics and structural biology.

Project 2 is funded by the University of Leeds in collaboration with Janssen Pharma­ceutica (Beerse, Belgium) and focuses on developing an understanding of how structured nucleic acids interact with lipids. Oligonucleotide-based drugs are of rapidly growing interest in the pharmaceutical industry, but current therapeutics remain limited beyond vaccines due to poorly understood delivery mechanisms.

Advanced formulations for oligonucleotide delivery such as lipid nanoparticles (LNPs) are frequently used, but the effect of lipids on the higher-order structure of therapeutics remain poorly understood.

You will have the unique opportunity to study the effect of how proteins and lipids interact with oligonucleotides and how this affects their higher-order structure, by pioneering mass spectrometry approaches for DNA/RNA biology.

For this role, you should have a PhD (or close to completion) in Biophysics, Chemistry, Structural Biology, Biochemistry or a related discipline with hands-on experience in structural mass spectrometry (particularly HDX-MS is desirable).
