本课题组致力于设计开发具有特定功能的人造蛋白材料,以满足当前资源、能源、环境等可持续发展策略对新材料的迫切需求。本实验室通过将生物物理、生物工程以及医学等领域进行交叉融合,建立了以蛋白功能为导向、以生物合成和多尺度模拟为基础、以高通量制备分析和功能测试为验证和优化手段的人造蛋白功能材料创制体系。主要研究方向为(1)蛋白质序列-结构-功能的关系以及蛋白质材料对人体组织/细胞生长分化的影响;(2)智能蛋白质材料设计、高通量筛选、蛋白质自组装理论研究和蛋白质材料物理化学改性;(3)蛋白质基柔性电子设备研发等应用方面的研究工作。

Dr. Huang’s lab focuses on predictive design, synthesis and functionalization of protein-based biomaterials for regenerative medicine and sustainable bio-electronics. Current research projects include: (1) injectable hydrogel and stem cell engineering, (2) stimuli-responsive biomaterial design, and (3) flexible electronics. Postdoc positions are available for highly motivated candidates with expertise in synthetic biology, material science and stem cell engineering.

本课题组通过多学科交叉,结合基因组学(RNA-seq, Bisulfite sequencing, ChIP-seq, ATAC-seq, Hi-C, single cell-based sequencing)、分子生物学以及生物信息学,来研究表观遗传在调控细胞 命运决定过程中的起到的在基础科学层面以及临床转化方面相关的问题。主要的研究方向包括:1)研究转座子在细胞命运决定过程中涉及的表观遗传调控相关问题。2)开发用于预测细胞命运的基于机器学习/深度学习相关的基因组学算法。3) 基于表观遗传组学的疾病分型以及疾病预测。

Dr. Chen’s lab focuses on the regulation of human embryogenesis and embryonic stem cell differentiation. Postdoc positions are available in Chen lab for the following projects

(1) The regulation of human embryonic stem cell differentiation at epigenetic and post-transcriptional levels.

(2) The function of RNA-binding protein during human primordial germ cell specification and differentiation.

(3) The new functions of cell fate determinant transcription factors during embryonic stem cell differentiation.

研究领域Research: The Leptihn Group offers two Postdoctoral Fellowships; one project focusses on the investigation of membrane proteins controlling cellular signalling pathways employing microfluidics and single molecule methods (fluorescence, electrophysiology) together with the University of Edinburgh and the ZJUI (Zhejiang University-University of Illinois Urbana Champagne). The second project involves viruses of microbes (bacteriophages) with aspects on both, function and structure of phage proteins, as well as phage therapy. Experience in imaging and protein biochemistry or microbiology are desired.

本课题组将从细胞器交流角度解析细胞-外界环境的相互作用,即外界环境变化(特指外源的能量供给变化)对细胞器互作结构特质(property/稳态(homeostasis)的影响。直接研究方向主要集中在两个方面:1)结合领域内已知和本人的前期数据,对细胞器互作网络改变细胞器膜脂组成和蛋白成分进行深入的机制研究。2)利用新的生物标记或技术从亚细胞器水平上发现细胞器互作网络对单个细胞器的影响,进一步拓展我们对细胞器互作网络的功能认知。

郭博士从事干细胞和癌症的研究,精于人类和小鼠遗传学,分子生物学,细胞生物学,生物医学,及基因组学的方法。他在造血发育、白血病干细胞以及肿瘤耐药调控方面等有重要建树。他建立了第一个由PTEN缺失导致T细胞急性白血病的小鼠模型,促成临床发现只带有PTEN突变而没有NOTCH1突变的婴幼儿T细胞白血病病人。他首次分离了T细胞急性白血病干细胞,揭示出了多因子突变导致白血病干细胞形成的过程,并发现了mTOR抑制剂的双重抗癌机制。近来,郭博士课题组从临床肺肿瘤中分离出肿瘤干细胞,并发现NOTCH1信号通路调控肺肿瘤干细胞的自我更新和铂类耐药。

Dr. Wei Guo laboratory uses basic biological approaches to understand biomedical challenges and translates its understanding to clinical practice. There are two major research directions: (1) stress response and ex vivo expansion of hematopoietic stem cells; and (2) regulation and targeted inhibition of cancer resistance.

研究方向:课题组运用分子生物学、细胞生物学、生物信息学、3D类器官培养和转基因小鼠模型研究干细胞命运控制,衰老及相关疾病的靶向治疗。

正在开展的课题:

1)肠干细胞稳态平衡、恶性转化及肠道疾病的早期诊疗。运用生物信息学分析、小鼠模型和肠道类器官,我们将深入研究大肠肿瘤干细胞的恶性转化和肿瘤转移的分子机制,并开展临床转化研究。

2)干细胞发育、衰老和融合的信号调控。实验室的最新成果揭示了首个永久性的阻断体内细胞融合的动物模型。在此基础上,我们将进一步探讨细胞融合与衰老的分子机理。

3)肌肉萎缩症,糖尿病和肥胖的靶向治疗。实验室已发表的研究证实了新的疾病治疗靶点。为此,我们将运用高通量筛选实验找寻靶点基因的小分子抑制剂,进行临床前期研究。

细胞通过感知和转导机械力信号来产生恰当的胞内生物化学应答。细胞内外的机械力都有助于胞质分裂的成功,从而使一个细胞分裂为两个子细胞。研究表明细胞胞质分裂的失败与基因组不稳定性、肿瘤进化和衰老紧密关联,而胞质分裂的成功则与细胞动力学和细胞外基质的相互作用密切相关。我们的研究目的是探究胞质分裂在衰老或其它病理状况下的对细胞外基质不同刚度反应的机械力学和动力学机制。为此,我的实验室联合采用化学遗传学、光遗传学和高分辨率活细胞显微成像技术探究胞质分裂失败产生的结果和胞质分裂的机械生物学机制。

Cells sense and transduce mechanical signals to generate appropriate biochemical and cellular response. Both the cellular and extracellular mechanics contribute to the success of cytokinesis, which is the division of one cell into two. Failure of cytokinesis is implicated in genomic instability, tumor evolution, and aging. The success of cytokinesis is linked to the proper interaction of cytokinetic machinery and the extracellular matrix. We aim to study the mechanics and dynamics of cytokinesis in response to different extracellular matrix stiffness during aging or other pathological conditions. To this end, my lab couple chemical genetics, optogenetics and high resolution live cell microscopy imaging to study the mechanobiology of cytokinesis and the consequences of its failure.

Leveraging CRISPR-mediated genome and epigenome engineering tools, NGS and bioinformatics, his lab aims for making prominent impact on stem cell study, cancer diagnosis and prognosis in a long run.

The centrosome is an important signalling and microtubule organizing centre in mammalian cells. Centrosomal defects are a common in high grade cancer and have been shown to trigger tumorigenesis in mice. The impact of centrosomal defects in deregulating the cell cycle in cancer is poorly understood. In addition, cancer genome sequencing efforts worldwide show mutations on centrosomal components are common. Using genetic, cell biology and biochemical assays, we aim to study how the changes in centrosomal signalling lead to the deregulation of the cell cycle in cancer.
