Both G protein coupled receptors (GPCRs) and Wnt proteins play important roles in a wide range of biological and pathophysiological processes. We are currently using various biochemical, molecular and cell biological, structural biological, genomic, proteomic, large scale RNAi screening, and transgenic and gene targeting approaches to: 1) characterize signaling pathways and networks initiated from GPCR ligands and investigate their roles in regulation of leukocyte functions (chemotaxis, infiltration, survival, reactive oxygen production, and degranulation) and in infection and inflammation-related diseases; 2) interrogating signaling mechanisms underlying fibroblast and tumor cell migration and their roles in tumor metastasis and fibrosis; and 3) elucidate Wnt signaling mechanisms and their functions in tumorigenesis and stem cell biology.
